DNA Fragmentation in Sperm

In order to complete a DNA fragmentation test in sperm it is necessary to provide a sperm sample in a sterile container.

Pre-requisites:

A seminogram of the sample (a morphological assessment is not necessary).

For the analysis of these samples it is better to fix the sample at the time it is obtained, so that the samples are sent already fixed. (If you would like the protocol for fixation, please contact the Genetics Laboratory at Institut Marquès).

One of the reactants needed to complete the fixation of the samples is 4% Paraformaldehyde in PBS 1X. Given that during the process of preparation of this solution extremely toxic fumes are released, the Genetics Laboratory at Institut Marquès can provide pre-prepared aliquots of this reactant so you can complete your fixation of the samples (the pre-prepared solution should be maintained at an room temperature.

For the first few samples sent from your laboratory it is necessary to check the fixed sample is reaching us safely, as good fixation is vital to create a viable result. To do this we shall request that you send us half of then sample fixed in your laboratory, and half sent fresh directly to the Genetics Laboratory at Institut Marquès, and then we shall conduct a parallel analysis.

Sending samples:

If the fixing process has not yet been tested:

  • Send the half of the sample that has been fixed in your Centre following the Genetics Laboratory at Institut Marquès’ protocol (keep the sample in the fridge until ready to send, and send it refrigerated).
  • The other half of the sample should be sent fresh and at room temperature. Please do NOT keep it in the fridge. From the time of collection of the sample to it arriving in our centre no more than 24 hours should have elapsed
  • All of the samples should be well sealed with parafilm stretch sealing film and the full name of the patient should be written on the outside in indelible pen.
  • Complete the request form for the study.
  • Send via email the name of the courier service used, and the number/code of the package to dgpi@institutomarques.com.

If the process of fixation has already been tested:

  • Following the protocol of the Genetics Laboratory at Institut Marquès, send the sample fixed in your Centre (keep the sample in the fridge until ready to send, and send it refrigerated).
  • All the tubes should be well sealed with parafilm stretch sealing film and the full name of the patient should be written on the outside with indelible pen.
  • Complete the request form for the study.
  • Send via email the name of the courier service used, and the number/code of the package to dgpi@institutomarques.com.

Results

The results will be sent 2 weeks after the sample arrives in the Genetics Laboratory at Institut Marquès. Once the results are obtained, these will be sent by encrypted email.
